In fact, themajority of PH domains bind PPIns promiscuously andwith low affinity in vitro and are diffuselylocalized in vivo. A novel class of PH domains -belonging to the OSBP/FAPP family - is identifiedhere with distinct phospholipid-binding properties,having both promiscuous and high affinity for PPInsin vitro. Additionally, thecrystal structure of amember of this class - Osh1p PH - has been determinedat high resolution. The PPIns-binding properties ofanother unusual PH domain, belonging to SH3BP2, isalso discussed, which has implications in the spatialand temporal regulation of 3-phosphoinositides.Finally, an alkylphospholipid drug is identified thatinteracts directly with the PH domain ofAkt1/PKBalpha in vitro, which has implications in theuse of PH domains as possible targets for therapeuticdrug intervention.
